Lady Jessica is Duke Leto's concubine, and also a member of the Bene Gesserit sisterhood. The BeneGesserit Sisterhood started 4,000 years before Dune to create a pure bloodline to produce the Kwisatz Haderach, a powerful messiah who could bridge time and space.
Duke Leto and Lady Jessica had a son against the Bene Gesserit's wishes. Their son is Paul Atreidies, the next Duke after his father, Leto.

Baron Harkonnen's nephew, Feyd-Rautha, was being trained by the Baron to eventually take over Dune to rule as a puppet ruler for the Baron.
One of the obsessions of Baron Harkonnen was Duke Leto's signature ring, a symbol of Duke Leto's power and authority on Caladan. During the capture of Arrakeen, Dr. Yueh took the ring from Duke Leto before he was brought to Baron Harkonnen and gave it to Paul as a memory.

The Mentat Master Assassins are a people who derive their speed and agility through the juice of Sapho, which speeds their mind and thoughts. The two Mentats from the movie are

and
Piter de Vries
The most recognisable feature of the Mentats are their red stained lips, a side effect from the juice of Sapho.
